Cell Line Collection: Doxycycline inducible NGN2 iPSCs across different MAPT genotypes
The ability to rapidly generate mature and functionally active iPSC neurons in a disease relevant background
has great potential to accelerate disease research. Here we have a collection of iPSC lines which combine
doxycycline-inducible Neurogenin 2 (NGN2) with different MAPT genotypes, enabling rapid generation of human neurons
while modelling tau-driven disease biology in a controlled, isogenic system.
These iPSC lines:
- Have an NGN2 cassette inserted at the AAVS1 locus for robust, inducible neuronal differentiation.
- Show rapid conversion to cortical-like neurons upon doxycycline treatment.
- Are from an isogenic background, differing only in MAPT genotype.
Cell line breakdown:
- SIGi001-A-15 carries iNGN2 in a MAPT wild-type background.
- SIGi001-A-17 carries iNGN2 alongside a MAPT homozygous exon10+16 variant.
- SIGi001-A-19 carries iNGN2 alongside a MAPT homozygous P301S + exon10+16 variant.
